ReliaPrep™ FFPE Miniprep Systems provide complete, all-inclusive methods for purification of gDNA and total RNA from formalin-fixed paraffin-embedded tissues.
Samples were analyzed using GoTaq® qPCR System. Earlier Cq values were obtained with the ReliaPrep™ FFPE Miniprep System.
More large-fragment RNA was purified with the ReliaPrep™ FFPE Total RNA Miniprep System than with competitor kits.
Panel A. 10μl purified gDNA used as template in multiplex PCR (GoTaq® Hot Start). Fragments ranging from 100-400bp detected.
Panel B. 5μl purified RNA used as template in GoScript™ Reverse Transcription System. 5μl of cDNA product used as template for the multiplex PCR (GoTaq® Hot Start). Fragments from 80bp-290bp detected.
All reactions performed in triplicate from sequential slices. 10μl PCR product run on a 2% agarose gel.
STR analysis was performed using Promega's Microsatellite Instability (MSI) Analysis System v. 1.2 (Cat.# MD1641). This system is compatible with STR amplification and MSI detection.
Sample Type: Human cervical cancer, 3 years old
Description: Genomic DNA was extracted using Qiagen (lane 1) and Promega ReliaPrep FFPE kits (lanes 2-5). Lane M = DNA marker, Lane N = blank.
Sample Type: Rat kidney, 2 years old
Description: Four 10µm FFPE sections were purified together (total amount = 40µm) and diluted in 40µl. Eight microliters were used for gel electrophoresis.
Sample Type: Human liver tumor, fixed less than 1 year
Description: One microliter out of 40µl total dilution of RNA was used for the gel.
Sample Type: Human kidney tumor, 2 months old
Description: Two samples were extracted using the total RNA kit. The total dilution volume was 30µl, and the loading volume for agarose gel electrophoresis was 15µl.
